Is it possible to configure Exchange 2007 to allow a user/mailbox to send messages with an arbitrary From address? Or do I have to define every address I want to send From as a Contact in AD first?

Turn on relay for authenticated clients (or based on IP address). Then make sure, if you aren't using SMTP authentication, to use "POP before SMTP" in your e-mail client.

This article might help if you are running 5.5 - http://technet.microsoft.com/en-us/library/dd277329.aspx

This is not related to SMTP, it's an Exchange "ADPermissions" issue.

And Exchange 5.5 is over 10 years old now, move with the times.
